Fructan Improves Survival and Function of Cryopreserved Rat Islets

Cryopreservation of pancreatic islets enables their long-term storage and subsequent transplantation; however, post-cryopreservation, viability, functions are reduced to a significant extent. Islet is composed five cells (? cell, ? ? ? PP cell), blood vessels that carry the nutrition. Freezing technology organization has not developed good method. This paper studied using fructan which been found effectively freeze protect material cell. transplantation established as an effective means treating patients with type 1 diabetes. In this study, we demonstrated effectiveness on cryopreserved by showing valid results for Isolated rat were phosphate-buffered saline (PBS) supplemented different concentrations and/or dimethyl sulfoxide (DMSO) in FBS. The survival rates estimated at time intervals, insulin secretion function was tested vitro. Furthermore, vivo syngeneic into streptozotocin-induced diabetic rats, grafts analyzed histologically immunohistochemically. Fructan significantly increased islet survival; 30% led more than 90% day 3, higher those DMSO groups (p < 0.05).
